Calbindins are Ca-binding proteins belonging to the troponin C superfamily. There are two types of CaBPs: the "trigger"- and the "buffer"-CaBPs. The conformation of "trigger" type CaBPs changes upon Ca2+ binding and exposes regions on protein that interact with target molecules, thus altering their activity. The buffer-type CABP are thought to control the intracellular calcium concentration. Parvalbumin (PARV, PVALB or oncomodulin) is a high affinity low mol wt (~12kD) cytoplasmic protein belonging to the troponin C superfamily. It is expressed in high levels only in fast-contracting muscles and at lower levels in brain and several endocrine tissues. PARV has two isoforms: alpha (rat/mouse/human: 110aa, 7p13-p11) and beta form (mouse/rat/human: 109aa) shares ~48% identity (mouse PARVs). The PARV-alpha is less acidic (pI=5) than the beta isoform and has an additional amino acid in the C-terminal helix. PARV-alpha is also expressed in inner ear. PARV-beta isoform is expressed in many fetal tissues, placenta and tumors.

Applications
Suitable for use in ELISA, Western Blot, Immunofluorescence and Immunohistochemistry. Other applications not tested.
Recommended Dilutions
ELISA: 1:10,000-1:100,000 using 50-100ng of control peptide/well. Reactivity is Calcium ion dependent. Western Blot: 1:2000-5000 (ECL). MW: ~12kD. Immunohistochemistry & Immunofluorescence: 1:500-1:1000 in indirect immunoperoxidase labeling of formalin-fixed, paraffin embedded sections of rat brain. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Purified frog muscle Parvalbumin
Form
Supplied as a liquid in PBS, 0.05% sodium azide

Specificity
Recognizes frog Parvalbumin. Crossreactivity with respect to the isoforms (PARV-alpha and beta) has not been studied. No significant crossreactivity is seen with other CABPs or EF-hand containing proteins. Species Crossreactivity: bovine, human, mouse, rat, canine, feline and fish
