The Apolipoprotein AI ELISA kit can be used to measure ApoAI in plasma and serum without prior sample purification. The standard curve spans the range of 0-20ng/ml with a limit of detection of less than 1.25ng/ml.
Principle of the Assay
This kit is based on a sandwich format. Each well of the microtiter plate has been coated with a monoclonal antibody specific of human ApoAI (capture antibody) and will bind any human ApoAI that is introduced into the well. A biotinylated detection antibody which binds selectively to a different epitope on the ApoAI molecule is then added to the well. After incubation and washing, excess reagents are washed away and avidin cholinesterase reagent is added to each well. The concentration of the analyte is then determined by measuring the enzymatic activity of the AChE by adding Ellman’s reagent (which contains the substrate for AChE) to each well. The product of the AChE-catalyzed reaction has a distinct yellow color which absorbs strongly at 412nm. The intensity of this colored, determined spectrophotometrically, is directly proportional to the concentration of ApoAI in the sample.

Storage and Stability
Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Kit is stable for at least 6 months.
