The protein encoded by this gene is a member of the Fe65 protein family. It is an adaptor protein localized in the nucleus. It interacts with the Alzheimer's disease amyloid precursor protein (APP), transcription factor CP2/LSF/LBP1 and the low-density lipoprotein receptor-related protein. APP functions as a cytosolic anchoring site that can prevent the gene product's nuclear translocation. This encoded protein could play an important role in the pathogenesis of Alzheimer's disease. It is thought to regulate transcription. Also it is observed to block cell cycle progression by downregulating thymidylate synthase expression. Multiple alternatively spliced transcript variants have been described for this gene but some of their full length sequence is not known.

FE65 is a signaling adaptor of APP protein. The FE65/APP complex was shown to translocate into the cell nucleus and regulate gene expression. Genetic studies in mice validated FE65-APP interaction and molecular studies provided evidence on the role of the complex in abnormal signaling detected in brains of Alzheimer Disease patients.
Applications
Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilution
ELISA: 1:128,000 Western Blot: 0.1-0.3μg/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a liquid in Tris-saline, pH 7.2, 0.5% BSA, 0.02% sodium azide.
Purity
Purified by immunoaffinity chromatography.
Specificity
Recognizes APBB1 at ~100kD in lysates of NIH/3T3 and HeLa (calculated MW of 77.2kD according to NP_001155.1 and 77.0kD according to NP_663722.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Zambrano et al, J Biol Chem. 1998 Aug 7;273(32):20128-33.; PMID: 9685356).
