Annexin A2 (ANXA2), is an ~40kD member of the Annexin family of calcium-dependent phospholipid-binding proteins that are preferentially located on the cytosolic face of the plasma membrane. Phosphorylation of Annexin A2 at Tyr23 regulates its involvement in endosomal trafficking and actin cytoskeleton rearrangement. The Annexins consist of a unique amino terminal domain followed by a homologous C-terminal core domain containing the calcium-dependent phospholipid-binding sites. The C-terminal domain is comprised of four 60-70aa annexin repeats. Annexin A2 also functions as an autocrine factor to enhance osteoclast formation and bone resorption and is a major cellular substrate of the tyrosine kinase Src. Human Annexin A2 shares 97% identity with mouse and rat Annexin A2.

Applications
Suitable for use in Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 0.1ug/ml Immunohistochemistry: 8-25ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recombinant corresponding to aa1-339 from human Annexin A2 expressed in E. coli (P07355).

Supplied as a lyophilized powder from PBS, pH 7.4, 5% trehalose. Reconstitute with 200ul sterile PBS.

Purified by Protein A/G affinity chromatography.
Specificity
Recognizes human Annexin A2. Species sequence homology: Recombinant human Annexin A1, A7, A9, A11, A13 (25%). No cross-reactivity with Annexin A3, A4, A6, A8, A10.
