A. fumigatus, a thermophilic, opportunistic and angio-invasive filamentous fungus, is the main causative agent of systemic bovine aspergillosis, a worldwide and often fatal respiratory disease of cattle. A3883-01C has been successfully used in immunohistochemistry for the specific and consistent in situ diagnosis of bovine systemic aspergillosis, attributed to its binding to the major cell wall component, galactomannan. A3880-01C has also been used for the identification of aspergillosis in human tissue sections.
Applications
Suitable for use in ELISA, Immunohistochemistry, Immunoprecipitation and Western Blot. Other applications not tested.
Recommended Dilutions
Immunohistochemistry (Paraffin): 1:300 Requires protein digestion pretreatment of paraffin sections. Western Blot: Detects a band of ~106kD of Aspergillus fumigatus wall fraction (WF). Optimal dilutions to be determined by the researcher.

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Purity
Purified by ammonium sulfate precipitation from tissue culture supernatant.
Specificity
Recognizes members of the Aspergillus spp. including A. flavus and A. niger, reacting strongly with walls and septae, and to a lesser extent within the cytoplasm of hyphae. Does not bind to water-soluble somatic antigens (WSSA) of Aspergillus spp., but may react with galactomannans of members of the genus Penicillium.
