Brain-derived neurotrophic factor (BDNF) is a member of the neurotrophin family of growth factors that includes NGF, NT-3, and NT-4. All neurotrophins have six conserved cysteine residues and share a 55% sequence identity at the amino acid level. BDNF has been shown to enhance the survival and differentiation of several classes of neurons in vitro, including neural crest and placode-derived sensory neurons, dopaminergic neurons in the substantia nigra, basal forebrain cholinergic neurons, hippocampal neurons, and retinal ganglial cells.1 BDNF is expressed within peripheral ganglia and is not restricted to neuronal target fields, raising the possibility that BDNF has paracrine or even autocrine actions on neurons as well as non-neuronal cells.2

Applications
Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested.
Recommended Dilutions
Immunohistochemistry (Paraffin): 8-25ug/ml using human spinal cord. Western Blot: 1-2ug/ml detects Brain Derived Neurotrophic Factor at ~300ng/lane under non-reducing and reducing conditions. Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Supplied as a liquid in PBS, pH 7.2, 5% trehalose.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes human Brain Derived Neurotrophic Factor. Will neutralize the biological activity of recombinant human Brain Derived Neurotrophic Factor. Shows <2% crossreactivity with recombinant human beta-NGF, recombinant human NT-3 and recombinant human NT-4 based on ELISA and Western Blot.
