Human CD235a (glycophorin A) is a sialoglycoprotein expressed early on the cell surface during erythropoiesis as pluripotent stem cells are recruited to differentiate into erythrocytes (1). CD235a (glycophorin A) is a useful marker to study the developmental biology of hematopoietic progenitor cells (2,3).

Applications
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilution
Flow Cytometry: 1:50. 80ul labels 10x10e6 human red blood cells. Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol
1. Wash 10x10e6 human red blood cells and preincubated 5 minutes with 20ul of 250ug/ml human IgG (to block non specific binding). 2. Incubated the cells for 45 minutes on ice with 80ul of C2548-98D3 at a 1:50 dilution. 3. Wash cells three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.08 log10 fluorescent units when compared to a Mouse IgM/FITC negative control at a similar concentration. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml anti-CD235a (Glycophorin A) antibody (C2548-98D1).
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
Immunogen
Neuraminidase treated human erythrocytes.
Form
Supplied as a liquid in PBS, pH 7.5, 0.2% BSA, 0.04% sodium azide, 5% glycerol. Labeled with fluorescein isothiocyanate (FITC).
Purity
Purified (90% SDS-PAGE).
Specificity
Recognizes an extracellular (peptide) domain of human CD235a.
