Geminin inhibits DNA replication by preventing the incorporation of the MCM complex into the prereplication complex (pre-RC). It is absent during the G1 phase, accumulates during the S, G2, and M phases, and disappears at the time of the metaphase-anaphase transition. Geminin contains 212 amino acids and has a destruction box sequence of RRTLKVIQP. Its destruction at the metaphase-anaphase transition permits replication in the succeeding cell cycle.
Applications
Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilution
ELISA: 0.05-0.2ug/ml Western Blot: 0.5-1ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute by adding sterile 40-50% glycerol, ddH2O. Reconstituted product is stable for 12 months at -20ºC. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
