IkBa protein is a NF-kB inhibitory protein. It binds NF-kB, retains the complex in the cytoplasma, prevents it from entering nuclear and functioning as a transcription factor. Upon receiving a variety of signals, IkBa undergoes phosphorylation at serine residues by a ubiquitin-dependent protein kinases and finally degraded by the proteasome. Free NF-kB then binds to DNA and affects gene expression.

Applications
Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation, Western Blot, Immunocytochemistry and Immunofluorescence Other applications have not been tested.
Recommended Dilutions
Flow Cytometry: 0.25-1ug/10e6 cells Immunoprecipitation: 1ug/ml Western Blot: 1-2ug/ml Immunohistochemistry: Paraffin sections Immunocytochemistry/Immunofluorescence: 1:100-1:1000 Optimal dilutions to be determined by the researcher.
Positive Control
Whole cell lysate from Jurkat, Daudi, or NIH 3T3.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recombinant protein corresponding to aa32-391 from human IkB alpha. Species Sequence Homology: rat
Form
Supplied as a liquid in PBS, 0.02% sodium azide.
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes both phosphorylated and non-phosphorylated forms of human IkB alpha at ~40kD. Species Crossreactivity: mouse
