The ion channels activated by glutamate are divided into two classes. Those that are sensitive to N-methyl-D-aspartate (NMDA) are designated NMDA receptors (NMDAR) while those activated by kainate and a-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid (AMPA) are known as kainate/AMPA receptors (K/AMPAR). NMDA receptors are among the most studied receptors in neuroscience because they are involved in neuronal cell development and plasticity, a cellular correlate for learning. NMDA receptors are also implicated in several disorders of the central nervous system including epilepsy and ischemic neuronal cell death. NMDA receptors also appear to be a target for ethanol at physiological concentrations and therefore may play a significant role in alcoholism.

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Western blot: 1:200 (NMDAR2B is present in high concentrations in the hippocampus) Immunoprecipitation: 3ul will (under appropriate conditions) quantitatively immunoprecipitate all NMDAR2B in 200ug of rat brain using solubilized hippocampal slices. Immunohistochemistry: Not suggested Note: Above dilutions are with 35S-protein A; with ECL dilutions may need to be considerably higher to obtain specific immunolabeling.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity
Recognizes rat NMDAR2B. By Western blot it recognizes a 180kD band in rat brain membranes. Also recognizes a 70kD band (thought to be a contaminant). No reactivity to NMDAR2A or NMDAR2C by Western blot. Immunolabeling is blocked by preadsorption with the C-terminal fusion protein that was used to generate the antibody. Species Reactivity: Human, rat, mouse and fish.
