p15INK4B is an ~15kD member of the p16INK4 family. Human p15INK4B is 138aa in length and contains four ankyrin repeats, a common structural sequence found in members of the INK4 family. The first 50aa of p15INK4B share 44% sequence identity with p16INK4. Following this is an 81aa region sharing 97% sequence identity, after which the two proteins diverge. Human p15INK4B shares 83% aa sequence identity with mouse and rat p15INK4B. Functionally, p15INK4B interacts strongly with CDK4 and CDK6 and is a potent inhibitor. p15INK4B can potentially induce cell cycle arrest by acting as an effector of TGF-B.
Applications
Suitable for use in Direct ELISA,, Western Blot and Immunocytochemistry. Other applications not tested.
Recommended Dilution
Western Blot: 1ug/ml using 293T human embryonic cell line and Hela human cervical epithelial carcinoma cell line Immunocytochemistry: 3-25ug/ml using immersion fixed HeLa human cervical epithelial carcinoma cell line Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
