Glucose M9Y medium, used to cultivate E. coli, is a minimal medium formulation supplemented with yeast extract. It is based on M9 salts with a pH of 6.9±0.2 and employs glucose as the carbon source. Alternative carbon sources may be substituted. Buffering is provided by a sodium-potassium phosphate system. Ammonium chloride provides a nitrogen source. Yeast extract is added to improve the protein production capacity of the medium and to permit cultivation of auxotrophic strains by supplying a source of amino acids, vitamins and minerals. This differs from the traditional formulation of glucose M9, which employs casamino acids. Our experience has shown that yeast extract typically gives higher yields of recombinant proteins than casamino acids. Glucose M9Y powder is supplied with a separate Glucose Nutrient Mix.

Our experiences with the expression of recombinant proteins have shown that the level of expression is very much dependent on the medium used. Some proteins are expressed well in one medium, but are not expressed in the same host strain in another medium or protein may be expressed in one medium while another protein is not expressed in the same medium.
This demonstrates why for each recombinant protein a medium screen becomes a crucial part of the optimization process. To facilitate the researchers’ efforts to develop the best systems for their proteins, we have developed a Media Optimization Kit.™. The kit is composed of six media formulations (including four that are unique and proprietary) which we have found over the years to be the most reliable for the increased production of recombinant proteins in Escherichia coli. The kit is used to quickly and easily identify the most suitable medium for the expression of a particular recombinant protein. Included are suggested approaches for improving the production of recombinant proteins, thus passing along the knowledge our staff scientists have acquired while working to produce a wide variety of proteins.

1. Dissolve15.5g per 1L dH2O. 2. Dispense into appropriate container. 3. Autoclave at 121°C for 15 minutes. 4. Aseptically add 20ml sterile Protein Expression Media, Glucose-Nutrient Mix, M9Y (Powder) pEX™ (P9102-40F). 5. Adjust pH to 7.2 ± 0.2.
Storage and Stability
Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
