Kit Component Usage
R1198-07A1: Raf 1 RBD, Recombinant, Human (Agarose), 1x600ug GST fusion-protein, corresponding to the human Ras Binding Domain (RBD, aa1-149) of Raf-1, expressed in E. coli. Form: Supplied as a 600ug of Raf-1 RBD agarose in 300ul of glutathione agarose beads. Provided as a 50% agarose gel slurry in PBS, 50% glycerol for a final volume of 600ul. Purity > 80%. Specificity: Specifically binds to and precipitates Ras-GTP from cell lysates. Species Crossreactivity: Predicted to cross-react with all mammalian species. R1198-07A2: Ras, p21, N,H,K Mab Mo xHu, 1x 100ug Isotype: IgG2aκ Immunogen: Purified recombinant Ras. Form: Supplied as a liquid in PBS, pH 7.4, 0.05% sodium azide, 30% glycerol. Concentration: ~1mg/ml Specificity: Recognizes p21 H-, K- and N-Ras. Species Crossreactivity: Human, mouse and rat. Note: This antibody may be reused 3-5 times for Iimmunoblot analysis. Add 0.05% sodium azide to the blocking buffer containing the diluted antibody. R1198-07A3: Mg2+ Lysis/Wash Buffer (5X), 2 x 18ml Form: Supplied as a liquid in 125mM HEPES, pH 7.5, 750mM sodium chloride, 5% Igepal CA-630, 50mM MgCl2, 5mM EDTA, 10% glycerol.
R1198-07A4: 10mM GTPgS (100X), 1 x 50ul Non-hydrolyzable analog of GTP. Form: Supplied as a liquid in 10mM GTPgS, sterile water. Quantity: Sufficient to label 5ml of cell lysates.
R1198-07A5: 100mM GDP (100X), 1 x 50ul GDP, Guanosine 5’-Diphosphate, type I, for in vitro labeling of G-proteins in the inactive form. Form: Supplied as a liquid in 100mM GDP, sterile water. Quantitity: Sufficient to label 5ml of cell lysates.
Storage and Stability
Store at -20°C. Stable for 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Quality Control Testing
R1198-07 precipitated Ras-GTP from HeLa cell lysates that had been loaded with GTPgS. The precipitated Ras-GTP was detected by Western blot using 0.05ug/ml R1198-07A2.
