In response to multiple cytokines and B and T cell receptor activation, the SH2 containing inositol 5' phosphatase, SHIP, becomes phosphorylated on tyrosine residues and associates with the tyrosine phosphorylated form of Shc. It may serve as a negative regulatory molecule under certain conditions, by preventing the activation of mast cells as well as B cell expansion, and by promoting apoptosis of confluent hemopoietic cells.

Applications
Suitable for use in Immunoblot and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Immunoblot Analysis: 0.5-2ug/ml Detects SHIP in RIPA lysates from human TF-1 cells (a bone marrow erythroleukemia cell line). TF-1 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S1013-20A (2ug/ml). Proteins were visualized using a goat anti-rabbit IgG labeled with HRP and a chemiluminescence detection system. Immunoprecipitation: 2ug Reported to immunoprecipitate SHIP from Ramos cells (human B cell line). Optimal dilution determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Fusion protein corresponding to aa7-133 of murine SHIP SH2 domain.
Form
Supplied as a liquid in 0.07M Tris, pH 7.4, 0.105M sodium chloride. No stabilizing proteins added. No preservative added. Also available with BSA & Azide. See S1013-20.
Purity
Purified by Protein A affinity chromatography.
Specificity
Recognizes SHIP, 145kD and/or 135kD as well as C-terminal truncated forms at 125kD and 110kD in some cell types. Species Crossreactivity: human and mouse.
