TNF-a, cachetin

TNF-alpha can be a useful tool to discriminate between the membrane form of TNF expressed on producer cells and the proteolytically cleaved, soluble TNF-alpha bound to its cognate cell membrane receptors (TNF-RI and TNF-RII). For this purpose we recommend to use this antibody in combination with the anti-TNF-alpha antibody, which recognizes soluble, membrane and receptor bound TNF-alpha.

Applications
Suitable for use in Flow Cytometry, Western Blot and FLISA. Other applications not tested.
Recommended Dilutions
Functional Assays: 1:50, used for inhibition of the biological activity of TNF-alpha. Western Blot: 1:50 Optimal ddilutions to be determined by the researcher.
Storage and Stability
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

BALB/c mice with huTNF, fusion of spleen cells with NSO myeloma cells
Form
Supplied as a liquid in PBS, 1% BSA, 0.02% sodium azide. Labeled with fluorescein isothiocyanate (FITC).
Purity
Purified by Protein G affinity chromatography.
Specificity
Recognizes the free soluble (17kD) and membrane (26kD) human TNF-alpha. The antibody inhibits the biological activity of both forms. It does not react with receptor bound TNF-alpha.
