NS3 and NS2b are non-structural viral proteins that play an essential role in viral replication and are therefore a potential target for treatment and prevention of West Nile Virus disease. NS3 is a trypsin-like serine protease that requires NS2b as a cofactor.
Applications
Suitable for use in Western Blot, Neutralization, Immunoprecipitation and Direct ELISA. Other applications not tested.
Recommended Dilutions
Western Blot: 2ug/ml using Recombinant Viral wnv NS3 Protease Neutralization: Measured by its ability to neutralize Recombinant Viral wnvNS3 Protease (0.5ug/ml) cleavage of the fluorogenic peptide substrate pERTKR-Amc (20uM). The Neutralization Dose (ND50) is typically 15ug/ml. Immunoprecipitation: 25ug/mL using conditioned cell culture medium spiked with Recombinant Viral wnvNS3 Protease Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized powder and reconstituted products must be stored at 4°C. For long-term storage, aliquot and store at 4°C. Do not freeze. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Ser1423-Lys1470, Gly1506-Leu1689 from wnvNS3 Protease, expressed in E coli.
Form
Supplied as a lyophilized powder in TBS, 5% trehalose. Reconstitute with 200ul sterile TBS.
Purity
Purified by Protein G affinity chromatography from hybridoma culture supernatant.
Specificity
Recognizes West Nile NS3 Protease.
