Wnt-9a (Wingless related integration site family member 9a, formerly Wnt-14) is a member of the Wnt family, which are ligands for frizzled family receptors. Wnt-9a is a secreted glycoprotein expressed in skeletal muscle and heart as well as in B-CLL and gastric, pancreatic and breast cancer cells. Human Wnt-9a is 98% aa identical with both mouse and rat Wnt-9a.
Applications
Suitable for use in Flow Cytometry and Western Blot. Other applications not tested.
Recommended Dilutions
Flow Cytometry: 2.5ug/10-6 cells MCF-7 human Breast cancer cell line fixed with paraformaldehyde and permeabilized with saponin. Western Blot: 5ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
Immunogen
Recombinant protein corresponding to Pro48-Glu92 and Arg231-Phe304 from human Wnt9a, expressed in E. coli.
Form
Supplied as a lyophilized powder in PBS, pH 7.4, 5% trehalose. Reconstitute with 200ul sterile PBS.
Purity
Purified by Protein G affinity chromatography from hybridoma culture supernatant.
Specificity
Recognizes human Wnt-9a. No cross-reactivity with rhWnt-2, -3a, -6, -7a, -7b, rmWnt-1, -2b, -4, -5a, -5b, -8a, -8b, -9b, -10a, -10b, -11 or -16.
