Ikaros family proteins, characterized by the presence of N-terminal zinc finger DNA-binding domain and C-terminal dimerization domain, belong to the Kruppel transcription factor superfamily. Ikaros family includes Ikaros, Aiolos, Helios, and possibly EOS and Pegasus (1). They can form homodimers as well as heterdimers with other members of the Ikaros family. Due to differential splicing, multiple isoforms can be generated and some behave in a dominant negative manner upon dimerization (2). Ikaros, also known as IKZF1 (Ikaros family zinc finger protein 1) and LYF-1 (Lymphoid transcription factor 1), is expressed abundantly in blood cells. Genetic studies in mice demonstrate that Ikaros is important for the normal development of B, T, natural killer, and dendritic cells, and that it functions as a tumor suppressor (3,4). Imbalanced expression of different Ikaros isoforms as well as deletions of Ikaros are associated with hematologic malignancies in humans (5-7).

Applications
Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilution
Western Blot: 1:1000. Incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to residues surrounding Pro329 of human Ikaros.
Form
Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol.
Purity
Purified by Protein A and peptide affinity chromatography.
Specificity
Recognizes endogenous levels of total human Ikaros protein. It is expected to recognize all isforms of Ikaros protein produced by differential splicing. Species Crossreactivity: mouse and rat
